Eine Vielzahl von Protein-Protein-Wechselwirkungen, die eine wichtige Rolle in Signaltransduktionsprozessen spielen, wird über Wechselwirkungen mit relativ kurzen Peptidsequenzen vermittelt. Dabei können vor allem über posttranslational modifizierte Proteinsequenzen Bindungen ausgelöst bzw. unterbunden werden. In der vorliegenden Arbeit wurden differentielle Peptid-Protein-Interaktionsexperimente (Pulldown-Experimente) mit phosphorylierten und den korrespondierenden unphosphorylierten Peptiden durchgeführt, um phosphorylierungsspezifische Interaktionspartner des Adapterproteins ADAP - als Teil des T-Zellsignalwegs und der adaptiven Immunantwort - zu bestimmen. Die Identifizierung der phosphorylierungsspezifisch interagierenden Proteine mit den ADAP-Peptiden erfolgte über quantitative Massenspektrometrie unter Verwendung von zwei Markierungsverfahren mit stabilen Isotopen: der Markierung von einzelnen Aminosäuren in Zellkultur (stable isotope labeling by amino acids in cell culture [SILAC]) und der enzymatischen 18O-Markierung mit schwerem Wasser.
